ABEL® Oxidative Stress Kits with Pholasin®

Test kits for oxidative stress research with a wide range of applications

Knight Scientific Limited (KSL) produces a range of unique oxidative stress test kits for measuring free radicals and other non-radical reactive oxygen containing species (ROS) produced by living cells and for determining the antioxidant potency of raw materials, ingredients and finished products. The antioxidant capacity of people and other species such as dogs, horses, pigs, fish and birds can also be measured. ABEL® kits have many applications:

There are two groups of kits, all of which are based on ABEL® (Analysis by Emitted Light) technology and contain the light-emitting protein PHOLASIN®, an ultra-sensitive chemiluminescent detector of free radicals and other reactive oxygen containing species (ROS):

  • Cell Activation Test Kits
  • Antioxidant Test Kits

KSL has many years experience of research involving oxidative stress and would be very pleased to help you decide on the most appropriate kit and experimental design. Just contact us and ask.

Customized kits and other pack sizes can be made to order to suit your needs

ABEL® Cell Activation Kits with Pholasin®

Test kits for whole blood or isolated cells

Chemiluminescent tests for measuring the real time production of free radicals and other non-radical reactive oxygen species (ROS) produced during the respiratory burst and degranulation of white blood cells (leucocytes). The assay has many applications:

  • Identify if cells have been activated.
  • Identify cells that may have been primed.
  • Show enhanced luminescence response to the receptor stimulant fMLP during infection.
  • Preview the way cells, circulating in the peripheral blood, are likely to respond at a site of inflammation.
  • Identify abnormalities in the leucocytes of so-called ‘normal’ control subjects.
  • Be used in drug assessment and discovery.
  • Measure superoxide to fmol concentrations.
  • Identify patients at greater risk of complications to surgical procedures or exposure to materials in medical devices such as those used in renal dialysis and cardiopulmonary bypass.
  • Monitor the progress of an inflammatory disease or complication and the response of a patient to drug intervention.

ABEL® Antioxidant Test Kits with Pholasin®

Chemiluminescent tests for measuring antioxidant capacity. The antioxidant capacity of the sample under test is expressed as the percentage reduction of peak luminescence of Pholasin® (and/or the delay in time in which the peak luminescence is observed) during the course of the test in the presence and absence of the sample. The results can be expressed against other antioxidant standards or as ABEL-RAC™ (relatively antioxidant capacity) scores per mg or per dose of material being tested.

  • Quantify antioxidant/prooxidant activity using a range of free radical challenges
  • Test raw materials, ingredients & finished products
  • Highly reproducible ABEL-RAC™ scores for labels of finished products
  • Identify possible synergy between ingredients
  • Optimise formulations
  • Monitor processes during manufacture
  • Confirm batch to batch uniformity
  • Quantify shelf life of ingredients and products

Samples of serum or plasma of people and other species are expressed as TAC (total antioxidant capacity) scores against a set of antioxidant standards of known concentration. These ABEL-TAC™ scores have known distribution ranges.

It is now possible to measure the antioxidant activity in a sample of blood taken from a mere finger prick to determine the TAC of individuals and classify their scores. These precise measurements make people aware of their antioxidant status, providing reassurance or enabling them to take steps to remedy a low value.

ABEL® 21 series: Antioxidant Test with Superoxide and Other Free Radicals

A chemiluminescent test for measuring the capacity of a sample of fluid such as water, plasma, serum, synovial fluid, food and food ingredients, cosmetics etc to scavenge free radicals such as the superoxide anion. An excellent test for screening antioxidants such as ascorbic acid (vitamin C).

ABEL® 31 series: Antioxidant Test with Halogenated Oxidants

A chemiluminescent test for measuring the capacity of a sample of fluid such as water, plasma, serum, synovial fluid, etc to scavenge halogenated oxidants such as hypochlorous acid and the longer-lived derived chloramines, produced naturally by some white blood cells. It is excellent for testing materials that are dissolved in organic solvents and assessing antioxidants such as albumin.

ABEL® 41 series: Wide-range Antioxidant Test with Peroxynitrite

A chemiluminescent test for assessing antioxidant capacity of a sample to scavenge peroxynitrite which attacks a wide range of antioxidants. This test is a total antioxidant capacity (TAC) test because peroxynitrite attacks virtually any substance that has antioxidant properties and can screen for both water and lipid soluble antioxidants. This assay is especially good for clinical studies aimed at assessing oxidative stress arising from excessive free radical production including peroxyl radical-mediated lipid peroxidation. It can also be used for assessing antioxidants in food, nutraceuticals, cosmetics and other media. Values of total antioxidant capacity are expressed in vitamin E analogue (VEA) units which can be used to compare individuals.

ABEL® 51 series: Antioxidant Test with Hydroxyl Radicals

A chemiluminescent test for measuring the capacity of a sample of fluid such as water, food extract, plasma, serum, etc to scavenge free radicals such as the hydroxyl radical. This is an excellent assay for assessing antioxidants in a variety of materials including plants.

ABEL® 61 series: Antioxidant and Superoxide Dismutase (SOD) Quantification Test

Chemiluminescent antioxidant test with enzyme generated superoxide produced in the xanthine/xanthine oxidase reaction. Excellent for quantifying superoxide and for measuring the activity of superoxide dismutase (SOD) and SOD mimetics. It is possible with this assay to distinguish between quenchers of superoxide such as Vitamin C and SOD mimetics.
